雌激素诱导小鼠肠道Cajal间质细胞内ERK快速活化的研究 被引量:11

摘要 目的研究17-β雌二醇(E2)对小鼠肠道Cajal间质细胞(ICC)中细胞外信号调节激酶(ERK1/2)的活化作用。方法利用原代培养小鼠ICC,采用Western blot分析E2诱导ICC内ERK1/2磷酸化的变化。结果E2作用于ICC,5 min即能诱导ERK1/2磷酸化,15 min后磷酸化ERK1/2蛋白表达达到最高,并从此逐渐降低;雌激素受体拮抗剂ICI182780可以阻断E2活化ERK1/2的作用。结论E2可以通过雌激素膜受体诱导ICC内ERK1/2的快速活化。 Objective To investigate the effect of 17-β estradiol(E2) on the activation of extracellular signal-regulated kinase (ERK1/2) in interstitial cells of Cajal(ICC). Methods Western blot were performed to examine the phosphorylation of ERK1/2. Results E2 rapidly induced phosphorylation of ERK1/2 within 5rain, E2-induced phosphorylation of ERK1/2 reached its peak at 15rain and rapidly declined thereafter. The effect of E2 on ERK1/2 activation were abolished by a selective estrogen receptor antago- nist,ICI182780. Conclusion E2 can induce rapid activiation of ERK1/2 in ICC through membrane estradiol receptors(mER).
