摘要
赤霉素(GA)与水稻矮化突变体的产生有密切的关系。根据GenBank数据库中水稻赤霉素代谢关键酶基因的cDNA序列,在保守序列区域设计引物,经过反应条件优化,建立了相对定量RT-PCR双标准曲线检测方法,并对中9B及其体细胞无性系矮化突变体SV9B的相关基因表达情况进行了检测。结果表明,该方法设计的引物特异性强,所构建的标准曲线相关性好,实验具有很高的可信度,可以应用于水稻赤霉素相关突变体的研究。
Gibberellin (GA) played an important part in generation of rice dwarf mutants. According to the eDNA sequences of rice gibberellin key metabolic enzyme genes from GenBank, primers were designed in conserved regions. With optimizing of reaction condition , a comparative quantitation RT-PCR detection method was established by two standard curve analysis. Detection was carried out on expression of such genes of Zhong 9B and SV 9B, whieh was the dwarf mutant of Zhong 9 B from somatic mutagenesis. The result demonstrated that the specification of primers was strong and the eorrelatability of standard curves was fine. The experiment had a high credibility, and could be promoted to the research of rice GA relative mutants.
