摘要
以生物素化羊抗兔免疫球蛋白和量子点标记链霉亲和素为荧光探针,采用竞争免疫分析模式,建立了一种灵敏度高、选择性好的检测雌三醇的荧光免疫分析方法。对几种测量参数如温育时间和温度、缓冲溶液pH、试剂浓度等进行了优化;方法的线性范围为0.01~1000ng/mL,检出限0.0029ng/mL,血清样品加标回收率在91%-117%之间。
A sensitive and selective method for the determination of estriol by fluorescence immunoassay (FIA) was established on the basis of competing mode immunoassay. The avidin-biotin system as signal amplification system and QDs-avidin probe were applied to the method. After the conditions of immunoreaction such as the time and temperature of incubating, the pH of the buffer solution and the concentration of the reagents optimized, the linear range and the detection limit of the method were 0.01 - 1000 ng/mL, 2.9 - 10^-3 ng/mL. The recoveries are in the range of 91% - 117% in human serum.
