丙戊酸钠抑制NB4细胞增殖及诱导凋亡的研究

摘要 目的:观察组蛋白去乙酰化酶抑制剂丙戊酸钠(VPA)对NB4细胞的作用。方法:台盼蓝细胞计数检测不同浓度VPA对NB4细胞增殖的抑制作用,应用流式细胞术检测VPA作用后NB4细胞CD11b的表达,用DNA梯形片段电泳分析细胞凋亡。结果:VPA能显著抑制NB4细胞的增殖(P<0.01),VPA处理NB4细胞72h时的半数抑制浓度(IC_(50))为1.17mmol/L;VPA组NB4细胞髓系分化抗原CD11b表达水平明显高于对照组(P<0.05);NB4细胞经不同浓度VPA处理48h后,出现典型的凋亡细胞所具有的梯形DNA条带。结论:VPA能抑制NB4细胞增殖,诱导细胞分化和凋亡。 Objective:To observe the effects of valproic acid sodium (VPA) on NB4 cells.Methods:Trypan blue cell counting was used to detect the effects of VPA in different density on proliferation of NB4 cells; Flow cytometry to observe CD11b expression of NB4 cells; DNA fragment ladder electrophoresis to observe apoptosis of NB4 cells.Results:VPA could obviously inhibit proliferation of NB4 cells (P 〈 0.01); The IC50 was 1.17 mmol/L treated with VPA for 72h. In VPA group, the CD11b expression of NB4 cells was significantly higher than control group (P 〈 0.05). NB4 cells appeared typical DNA ladder of apoptosis 48h after being treated with VPA. Conelusions:VPA can inhibit proliferation, induce differentiation and apoptosis of NB4 cells.
