摘要
目的建立HBV感染的急性乙型肝炎大鼠模型。方法将经磷酸钙沉淀的含3.2kb全序列HBVDNA的HBV-PCNCX质粒直接导入大鼠肝脏。结果10只实验动物中,8只大鼠血清HBsAg和HBeAg阳性,肝细胞中HBVmRNA及HBsAg得到表达,肝脏出现局灶性炎性细胞浸润,肝细胞坏死、浊肿等典型的病毒性肝炎病理变化。结论将磷酸钙沉淀后的HBVDNA导入大鼠肝脏细胞。
Objective The objective of the study was to estabolish an appropriate animal model of infection of hepatitis B virus. Methods HBV PCNCX plasmid with 3.2kb HBV DNA were introduced into rat livers after calcification with calcium phosphate. Results HBsAg and HBeAg were positive, as well as the serum glutamic pyruvic transaminase were elevated in 8 of 10 rats. The mRNA of HBV were detected by the technology of in situ hybridization. Focal infiltration of lymphocytes was observed in the livers. Conclusion Transfection of HBV DNA into rats liver resulted in hepatic histological and serological changes comparable to those of HBV induced acute hepatitis in human.
