摘要
目的:研制大鼠肺气虚证模型,为研究因慢性呼吸道感染导致肺气虚证的诊治提供可靠的实验动物。方法:健康SD大鼠随机分为空白对照组、烟熏对照组,以及高、中、低菌量模型组。模型组采用呼吸道生物被膜菌(bacterial biofilm,BF)铜绿假单胞菌滴鼻法,辅以寒冷疲劳刺激引起大鼠呼吸道慢性感染,诱导肺气虚证的发生。观察各组证候表现、游泳时间、体重变化、血液流变学指标的改变,以及肺支气管组织器官病理改变。结果:高、中、低菌量模型组大鼠的一般体征均符合肺气虚证,症状较烟熏组稳定。游泳时间和体重增加低于空白对照组,尤以高菌量组为明显(P<0.05,P<0.01)。血流变指标提示血液流变性有障碍,肺及支气管炎症明显,炎症的程度和感染菌量有关。结论:将呼吸道生物被膜菌(铜绿假单胞菌)以滴鼻法并辅以寒冷疲劳刺激,可以诱导大鼠肺气虚证模型,相关的指标表明证候表现与中医学理论的病机相符,且该模型具有稳固、易复制、贴近原型的特点。
To develop the rat model of lung-qi deficiency pattern so as to provide reliable animal model for the study of lung-qi deficiency pattern caused by chronic respiratory infections. Methods : Healthy SD rats were randomly divided into blank control group, smoked control group, high-, middle-, and low-concentration of bacterial model groups. The model groups were dripped with respiratory tract biofihn bacteria (Pseudonwnas aeruginosa) through noses combined with cold and fatigue stimu- lus to induce chronic respiratory infections and lung qi deficiency pattern. The symptoms, swimming time, changes in weight, blood rheology and pathogenesis in bronchial and lung tissues and organs were observed. Results : The general signs of the model rats were in line with tung-qi deficiency pattern of traditional Chinese medicine, and their symptoms were more stable than that of the smoked eontrol group. The swimming time and weight gain wm~ less than that of the blank control group, especislly for the group of high concentration of baete^ia(P 〈 0.05 or P 〈 0.01 ). Blood rheology was abnormal, bronchial and lung inflammation were evident, and the degree of inflammation was related to bacterial concentrations. Conclusion : Combined with droping respiratory tract biofihn bacteria (Pseudomonas aeruginosa) to the noses and cold and fatigue stimulus can develope the rat model of lungqi deficiency pattern. The related index indicates that the models are of the pathogenesis characteristic of traditional Chinese medicine. Besides, the model was stable, easy to be replicated, and simliar to clinical origin.
