摘要
目的观察益心口服液预处理对心肌缺血大鼠的影响并初步探讨其作用机制。方法利用Langendorff离体心脏灌注装置,结扎Wistar大鼠冠状动脉左前降支建立心肌缺血模型。将大鼠随机分为假手术组、模型组、地尔硫组、复方丹参滴丸组,以及益心口服液大、中、小剂量组,观察各组结扎前后心脏血流动力学的改变,测定各组心肌超氧化物歧化酶(SOD)的活性和丙二醛(MDA)的含量,检测各组心肌组织中腺苷酸(ATP、ADP、AMP)含量以及能荷(EC)的改变。结果①不同浓度的益心口服液预处理均能改善结扎导致的左室收缩压(LVSP)的降低(P<0.05);大、中剂量益心口服液在个别时间点上明显减缓了结扎导致的左室舒张末期压(LVEDP)的抬高(P<0.05),但对冠脉流量(CF)的改善作用不明显。②益心口服液大、中剂量预处理减轻了缺血心肌SOD活性的降低和MDA含量的升高(P<0.05),其作用与地尔硫组和复方丹参滴丸组无显著性差异(P>0.05)。③益心口服液大、中剂量和复方丹参滴丸均能缓解缺血心肌组织中ATP含量的衰减(P<0.05)。结论益心口服液预处理可对抗离体大鼠心肌缺血损伤,且有剂量依从性,其保护作用可能涉及到抗氧化损伤以及对能量的有效保存。
Objective To investigate the effects of "Yixin Oral Liquid" (YOL) preconditioning on myocardial ischemic rats in vitro. Methods The model of myocardial ischemia was established in rats by left coronary artery ligation for 120 minutes by means of Langendorff apparatus, and then the rats were divided into 7 groups randomly: sham operation group, model group, diltiazem hydrochloride group (TE), Fufang Danshen Dripping Pills group (FD), and various concentration of YOL groups (high, medium and low doses). The hemodynamic parameters (LVSP, LVEDP, and coronary flow ), superoxide dismutase(SOD) activity and malonaldehyde (MDA) content, and ATP, ADP and AMP levels in heart tissue were observed. Results Various concentrations of YOL preconditioning could obviously decrease LVSP(P 〈0.05 ), but improve CF little; the YOL groups (high and medium doses) could reduce SOD activity and increase MDA contents (P 〈0.05 ), with no significant differences in comparison with FD and TE group( P 〉0.05 ) ; the high and medium doses of YOL and Fufang Danshen Dripping Pills could raise ATP levels in ischemic heart tissue ( P 〈 0.05 ). Conclusion The YOL preconditioning contributes to protective effect with a dose-dependent manner on acute myocardial ischemic rats in vitro, and its effects of antioxidative stress and maintenance of ATP might be involved in the actions of YOL preconditioning.
