雷公藤多甙对小鼠异种骨移植急性排斥反应的抑制作用 被引量:1

摘要 目的:观察雷公藤多甙对小鼠异种骨移植急性排斥反应的抑制作用。方法:BALB/C小鼠随机分为治疗组(n=20)和对照组(n=20),治疗组小鼠行雷公藤多甙(30 mg.kg-1.d-1)灌胃3 d,对照组给予等量生理盐水灌胃,将猪骨作为供体分别移植至治疗组和对照组小鼠体内,移植2周后,通过流式细胞术检测小鼠脾脏中CD4+、CD8+T淋巴细胞数量,ELISA法检测2组小鼠血清中白细胞介素2(IL-2)、γ-干扰素(IFN-γ)和白细胞介素4(IL-4)的含量,观察移植骨周围组织形态学改变,比较2组小鼠异种骨移植后发生急性排斥反应的情况。结果:骨移植2周后治疗组小鼠脾脏中的CD4+、CD8+T淋巴细胞数量、CD4+/CD8+T淋巴细胞的比值较对照组均明显降低(P<0.05),治疗组小鼠血清中IL-2、IFN-γ和IL-4含量较对照组均明显降低(P<0.05),治疗组较对照组小鼠移植骨周围组织中淋巴细胞浸润明显减少。结论:雷公藤多甙可以减轻小鼠异种骨移植急性排斥反应。 Objective To investigate the effect of tripterygium wilfordii hoff-F on acute immune rejection of bone xenograft in mice. Methods Male Neijiang swine and BALB/c mice were used as donor and receipts, respectively. Forty mice were divided into experimental group and control group at random. The mice in experimental group were administered intragastrially with tripterygium wilfordii hoof-F before bone xenograft. At two weeks postoperatively, CD4^+ , CD8^+ , CD4^+/CD8^+ T lymphocytes in spleen were detected by flow cytometry, and the levels of IL-2, IFN-γ and IL-4 in blood were analyzed by quantitative ELISA assay. And the infiltration of lymphocytes and macrophages surrounding soft tissues of bone graft was also observed in order to evaluate the immunosuppression of tripterygium wilfordii hoof-F. Results At two weeks postoperatively, compared with control group, CD4^+ , CD8^+ , CD4^+/CD8^+ T lymphocytes in spleen, the levels of IL-2, IFN-γ and IL-4 in blood of mice in experimental group were significantly decreased (P〈 0. 05). There was few lymphocytes infiltration surrounding soft tissue of bone graft in experimental group. Conclusion Tripterygium wilfordii hoff-F can relieve the acute immune rejection of bone xenograft in mice.
