摘要
目的:探讨他汀类药物对人外周血T淋巴细胞的抗炎机制。方法:体外培养人外周血T淋巴细胞,用不同浓度的辛伐他汀进行处理,实时荧光定量PCR法检测T细胞肿瘤坏死因子-α(TNF-α)、白细胞介素-2(IL-2)mRNA表达水平,免疫细胞化学S-P法检测T细胞核因子-kappaB(NF-κB)移位情况。结果:辛伐他汀呈剂量依赖性抑制TNF-α、IL-2mRNA表达,抑制NF-κB移位入核。结论:辛伐他汀可对人外周血T淋巴细胞有抗炎作用,且可能是通过抑制NF-κB的转位入核实现的。
Objective: To observe the anti-inflammatory mechanisms of simvastatin in human T lymphocyte. Method:Jurkat cells were treated with different dose of simvastatin, the levels of IL-2 and TNF-α mRNA were quantified with real time fluorescent quantitative polymerase chain reaction(RQ-PCR), the transportion of NF-κB into the nuclear was detected by immunocytochemistry. Result: Simvastatin could significantly inhibit the production of IL- 2 and TNF-α( P 〈 0.05), and the transportion of NF-κB into the nuclear was inhibited by simvastatin (P 〈 0,05). Conclusion:simvastatin had the anti-inflammatory effect on T lymphocyte by inhibiting the transportion of NF-xB into the nuclear.
