摘要
目的观察雌激素缺乏对大鼠海马CA1/CA2区及额叶皮层海马胆碱能神经刺激肽(HCNP)含量的影响。方法选取清洁级4月龄雌性SD大鼠30只,随机分为3组:假手术组(Sham组)、去卵巢组(OVX组)和去卵巢加雌二醇(E2)组(OVX+E2组),每组10只。除Sham组做假性手术外,其余2组均行双侧卵巢切除术;OVX+E2组去卵巢手术10d后给予E2皮下注射,100μg/kg,隔日1次。去卵巢手术后常规喂养4个月,获取组织标本,用于组织蛋白和组织总RNA的提取。用Western印迹法测定大鼠海马和皮层C-HCNPpp、N-HCNPpp的相对含量。用RT-PCR技术检测HCNPpp mRNA水平。结果OVX组大鼠海马和皮层中C-HCNPpp的相对含量较Sham组和OVX+E2组明显升高(P<0.05),而Sham组和OVX+E2组之间无显著差别(P>0.05),三组大鼠之间的海马和皮层中N-HCNPpp的相对含量差异不显著(P<0.05);OVX组和OVX+E2组大鼠HCNPpp mRNA表达与Sham组无显著差异(P>0.05),OVX+E2组大鼠HCNPpp mRNA表达与OVX组相比增高(P<0.05);三组大鼠皮层HCNPpp mRNA表达无显著差异(P>0.05)。结论雌激素缺乏可导致大鼠海马CA1/CA2区及额叶皮层HCNP的增多。
Objective To observe the effect of the defect of estrogen on the contents of HCNP in regions of CA1/CA2 of hippocampus and the expression of HCNPpp mRNA in prefrontal cortex of ovariectomized (OVX) SD rats. Methods Thirty female clean SD rats aged 4 months were randomly divided into sham, OVX, OVX and estradiol (OVX + E) groups, and ten rats per group. Sham group was ablated some adipose tissue above ovaries, OVX group was ablated two sides ovaries, OVX + E group was supplied by estradiol(100μg/kg, qod) after 10 d of ovariectomy. Postoperative rats were fed by routine method and then sacrificed 4 months later to obtain tissues and specimen for the use of the extraction of tissues' protein and total RNA. The relative protein contents of C-HCNPpp and N-HCNPpp were measured by Western blotting, and the expression of HCNPpp mRNA was measured by RT-PCR in the dorsal hippocampus and prefrontal cortex of ovafi- ectomized rats. Results Compared with that of sham group and OVX + E group,the relative content of C-HCNPpp of OVX group in hippocampus and prefrontal cortex was higher, and there was not different between sham group and OVX + E group(P〉0.05). However, the expression of N-HCNPpp in hippocampus and prefrontal cortex was not different among three groups(P〉0.05). Compared with that of sham group, the relative expression of HCNPpp of OVX group and OVX + E group in hippocampus was not different (P〉0.05). Compared with that of OVX group, the relative expression of HCNPpp mRNA of OVX + E group in hippoeampus was higher (P〈0.05). The relative expression of HCNPpp mRNA in prefrontal cortex was no different among three groups(P〉0.05). Conclusions The deficiency of estrogen in female rats leads to the increasing of the relative amount of HCNP in CAI/CA2 of hippocampus and prefrontal cortex of ovariectomized rats.
