谷氨酰胺对脓毒症大鼠基质金属蛋白酶及血栓调节蛋白基因表达的影响 被引量:1

摘要 目的观察脓毒症大鼠血浆基质金属蛋白酶(MMP-9)、血栓调节蛋白(TM)浓度和TM基因表达的变化及研究谷氨酰胺(Gin)对其影响,以明确Gin对脓毒症大鼠内皮细胞的保护作用。方法采用内毒素(LPS)腹腔注射法制作脓毒症大鼠模型,大鼠随机分为对照组、LPS组及LPS+Gin组,每组再分为0、6、12、24h亚组(n=6),检测各时点血浆MMP-9及TM的浓度及心肌细胞TMmRNA的表达。对结果进行方差分析。结果本研究显示,与对照组比较,在脓毒症后LPS组、LPS+Gin组血浆MMP-9、TM的浓度逐渐升高,至12h达到峰值,24h有所下降(P〈0.05),但LPS+Gin组升高幅度较LPS组明显减小(P〈0.05)。LPS组、LPS+Gin组在术后TMmRNA表达水平均不同程度高于对照组,12h达到高峰(P〈0.05),然后下降;LPS+Gin组升高幅度较LPS组明显下降(P〈0.05)。结论脓毒症增加大鼠血浆MMP-9及TM含量及上调TMmRNA基因的表达;脓毒症大鼠应用Gin进行干预治疗能够降低MMP-9及TM含量及抑制脓毒症早期组织的TMmRNA表达;Gin对脓毒症血管内皮细胞具有保护作用。 Objective Studies the change of matrix metalloproteinase - 9 ( MMP - 9 ) and thrombomodulin(TM) in rats with sepsis and effect of glutamine (Gin) on them; investigate the protective effect of Gin on endothelial cell after sepsis. Methods Sepsis rat models were finished through peritoneal injection with lipopolysaccharide, and divided into control group, LPS group and LPS + Gin group, which were divided into four subset groups, 0, 6, 12, 24 h ( n = 61) The change of serum concentration of MMP -9,TM and expression of myocardial TM mRNA were examined. The dates were tested statistically by F - test. Results Compared with control group, The serum level of MMP - 9 and TM in LIPS group and LPS + Gin group were higher after sepsis ( P 〈 0. 05), and is the highest at 12 h (P 〈 0.05 ). LPS + Gln group was significantly decreased than LPS group (P 〈0. 05). The expression level of TM mRNA is gradually rose after sepsis in myocardial cell in LPS group and LPS + Gin group, and is most at 12 h ( P 〈 0. 05). The level of TM mRNA in LPS +Gln group is the lower than one in LPS group(P 〈0. 05). Conclusions Sepsis increases the plasma level of MMP - 9 and TM, and has effect of high - regulating the expression of TM mRNA in rat. Gln can decrease the level of MMP - 9 , TM and the expression of TM gene. So that Gin can protect the vein endothelial cells of rat with sepsis.
