摘要
以黄花马蹄莲块茎作为外植体,从灭菌的时间和条件,不同激素含量的搭配,以及黄花马蹄莲的生长时间、外植体自身大小等因素,针对组培的前期环节改进和提高丛生芽的诱导率进行研究。结果表明:初代体系的建立以500倍50%多菌灵稀释液处理4 h,0.1%升汞溶液处理13 min污染率最低,将芽块切为直径0.4~0.6 cm的大小,接种至MS+NAA 0.1 mg/L+6-BA2 mg/L的诱导培养基上效果最好。使用不同生长时期彩色马蹄莲块茎进行诱导在前期的差异较大,经后期继代后差异减小。
Zantedeschia elliottiana tubers were cultured on MS medium as test materials. The research was concerned with different factors which including sterilization times and conditions, contents of hormone, growth times of plants and sizes of explants to improve the prophase link in tissue culture and raises the induetivity of adventitious buds. The result indicated that: immerse the tuber in 50% carbendazim by 500 times water in 4 hours and then in 0. 10% mercury bichloride solution in 13 minutes had the lowest rate of pollution in primary culture. Tubers about 0. 4-0. 6 cm in diameter on MS+NAA 0.1 mg/L+6-BA 2 mg/L had the best effects on inducing caespitose shoots. Tubers from different growth times of Zantedeschia elliottiana had different inductivities in primary culture, but the differences reduced after several succesive transfer cultures.
