醋酸水解三七总皂苷制备人参皂苷Rg3工艺的优选 被引量:12

Process optimization of ginsenoside Rg3 prepared by hydrolysing Panax Notoginseng saponins by acetic acid

摘要 目的:寻找醋酸水解三七总皂苷制备人参皂苷Rg3(Ginsenoside Rg3)的最佳工艺。方法:考察醋酸体积分数、水解温度、皂苷体积分数、水解时间4个因素对人参皂苷Rg3产量的影响;在单因素试验的基础上进行L9(34)正交试验,以总水解产物[主要含20(R)-人参Rg3]的质量为考察指标。结果:醋酸水解三七总皂苷制备人参皂苷Rg3的最佳工艺条件为:醋酸体积分数30%、水解温度90℃、皂苷体积分数5%、水解时间1h;本试验所确定的最佳制备工艺制备人参皂苷Rg3的得率为30%,HPLC法检测总转化产物中20(R)-人参皂苷Rg3含量达80%以上,旋光分析其光学纯度为78.33%。结论:该法是一种省时、省力、简便、快速制备较高纯度的20(R)-Rg3的可行的方法。 OBJECTIVE To find the best process of ginsenoside Rg3 preparated by acetic acid hydrolysing panax notoginseng saponins. METHODS The influencing factors were investigated: acetic acid concentration, temperature, saponin concentration and time; basing on the single factor experiment, the L9 (3^4 )orthogonal test was prepared. The quality of ginsenoside Rg3 from hydrolysate was used as index,and the experiment was repeated three times. RESULTS The best process was as flows: the acetic acid concentration was 30%, the temperature was 90 ℃, the saponin concentration was 5%o and the time is three hours; the yield of 20(R)-ginsenoside Rg3 prepared by this process was 30% calculated by the conversion of panaxadiol sapo nins. and optical purity was 78.33%. CONCLUSION The method is easy to operate and is a feasible method to prepare high purity 20(R)-ginsenoside Rg3.
