摘要
建立测定马蹄皮中总黄酮含量的光谱分析方法。以40%乙醇为溶剂,微波辅助提取马蹄皮中的总黄酮。以芦丁为标准样品,以NaNO2-Al(NO3)3-NaOH为显色剂,采用可见分光光度法在510nm测量波长测定马蹄皮中总黄酮含量。回归方程A=0.0105C+0.0014,相关系数R2=1,黄酮浓度在8.41mg/L~82.88mg/L范围内服从比耳定律。测定结果显示,马蹄皮中总黄酮的平均含量为1.59%,重现性试验RSD=1.38%(n=6),平均回收率99.6%(RSD=0.799%,n=6),精密度试验RSD=1.64%(n=6)。
A method for the determination of total flavonoids from water chestnut peel was established. With 40 % ethanol as extracting solvent, the total flavonoids were extracted from water chestnut peel by microwave. With rutin as standard sample and NaNO2-Al (NO3)3-NaOH as color reagent, the flavonoids content was determined by VIS-spectrophotometry at the wavelength of 510 nm. The regression equation was A=0.010 5 C+ 0.001 4, correlation coefficient R^2=1, linear range of obeying Bill Law was 8.41 mg/L-82.88 mg/L of total flavonoids concentration. The determination result showed that the average content of total flavoniods from water chestnut peel was 1.59 %, the RSD of reproducibility test was 1.38 %(n=6). The average recovery of this method was 99.6 % (RSD=0.799 %, n=6), the RSD of precision test was 1.64 %(n=6).
