摘要
目的研究白念珠菌对人外周血单核细胞源树突状细胞(dendritic cells,DCs)形态及表面分子的影响。方法用科玛嘉念珠菌显色培养基及芽管试验分离、鉴定白念珠菌;体外培养正常人外周血单核细胞源DCs,经不同剂量的白念珠菌、无菌生理盐水作用后,在倒置显微镜下观察细胞的形态变化,用流式细胞仪检测表面分子的表达。结果实验组与对照组DCs细胞形态无差异;实验组DCs表面分子CD80、CD86表达明显高于同等剂量对照组,差异有显著性意义(P<0.05),且表达量与白念珠菌剂量呈正相关(P均<0.01)。结论在一定剂量范围内,白念珠菌可促进DCs表面分子CD80和CD86表达,DCs进一步成熟。
Objective To investigate the effects of candida albicans on the surface molecules and the shapes of the human peripheral blood monocyte-derived dendritic cells. Methods Candida albicans were separated and judged by the CHROM agar Monilia colouration nutritive medium and the germ-tube-formming tests. The dendritic cells were obtained by culture in vitro. Then after stimulated by the different doses of candida albicans suspension or germfree isotonic Na chloride, shapes of the cells were observed under the inverted microscope and the expressions of CDs0 and CD86 were analyzed by FACS. Results The expressions of CO80 and CD86 in the experimental groups were much higher than those in the same dose of control groups( P 〈 0. 05 ) . Furthermore the levels was correlated to the doses of candida albicans directly. But no changes of the shapes of the cells were detected in the two groups. Conclusions In the definite doses, candida albicans can enhance the expressions of CD80 and CD86, which enhance the maturation of the dendritic cells.
