系统性红斑狼疮患者Th_1/Th_2转录因子基因表达失衡的分析

Analysis of the Gene Expression Imbalance of Th_1/Th_2 Associated Transcription Factors in Patients with Systemic Lupus Erythematosus

摘要 目的检测T-bet和GATA-3的基因表达水平以及调节性T细胞相关细胞因子水平,探讨转录因子对调节性T细胞相关细胞因子水平的影响及其在系统性红斑狼疮(SLE)发病中的作用。方法利用实时荧光定量PCR技术检测SLE患者及正常人外周血单核细胞(PBMC)中T-bet和GATA-3mRNA的表达状况;用酶联免疫吸附试验(ELISA)检测SLE患者及正常人血清中IL-10和TGF-β水平。结果SLE患者T-betmRNA表达低于正常人,但GATA-3mRNA表达明显高于正常人,差异均具有统计学意义(P均<0.05)。SLE患者血清中IL-10明显高于正常人,差异具有统计学意义(P<0.05);两组的TGF-β水平比较,差异无统计学意义(P>0.05)。结论外周血单核细胞GATA-3mRNA表达增高及T-betmRNA表达降低,导致SLE患者存在Th1/Th2细胞免疫的不平衡,调节性T细胞相关细胞因子IL-10水平升高,TGF-β没有明显变化,其均导致SLE患者体内免疫系统的紊乱。在SLE发病中起重要作用。 Objective To investigate the role of transcription factors in treg cells related cytokines and the pathogenesis of systemic lupus erythematosus (SLE) , we analyzed the gene expression levels of T-bet and GATA-3 and treg cells related cytokines. Methods The expressions of transcription factors T-bet and GATA-3 in PBMC from patients with SLE and the healthy subjects of control were detected by fluorescent real-time quantitative PCR, the levels of IL-10 and TGF-β in serum from patients with SLE and the healthy subjects in the control group were detected by ELISA. Results T-bet mRNA expression in patients with SLE was lower than that among the subjects in the control group, and the difference was statistically significant( P 〈 0.05 ) ; GATA-3 mRNA expression in patients with SLE was higher than that of the control group, the difference was found to be statiscally significant( P 〈 0.05 ). The quantity of IL-10 in serum of patients with SLE was significantly higher than that in the healthy subjects of the control group( P 〈 0.05 ) ;and there was no significantly difference in TGF-β between the two groups (P 〉 0.05 ). Conclusions That the gene expression level of T-bet was decreased and GATA-3 was increased in PBMC of SLE induces the cell mediated immunity imbalance between Th1/Th2, the level of IL-10, treg cells related cytokines, increased, TGF-β have no marked change. These activities cause the disorder of the internal immune system of the patients with SLE and play an important role in SLE development.
