盐酸千金藤碱对鼻咽癌SUNE-1细胞生长抑制的观察 被引量:3

Inhibitory effect of cepharanthine hydrochiorid on human nasopharyngeal carcinoma cell line SUNE-1 in vitro

摘要 目的:研究盐酸千金藤碱(CH)对鼻咽癌细胞系SUNE-1的生长抑制和诱导凋亡作用。方法:采用MTT法检测CH对SUNE-1细胞生长的抑制作用,分别用流式细胞分析仪、透射电镜观察CH处理SUNE-1细胞后细胞周期阻滞和细胞凋亡的发生。结果:MTT法证实,CH对SUNE-1细胞生长有抑制作用,且呈剂量-效应关系和时间-效应关系,P<0.01。流式细胞仪检测发现,CH处理SUNE-1细胞后,实验组G0/G1细胞(71.5±8.7)%明显高于对照组(46.9±10.5)%,P<0.05,提示细胞周期停滞于G0/G1期,流式细胞周期分布图可见凋亡峰。透射电镜可见,SUNE-1细胞出现凋亡细胞形态学改变。结论:CH对SUNE-1细胞有抑制作用,机制可能与阻滞细胞周期及诱导细胞凋亡相关。 OBJECTIVE: To study the inhibitory effect of cepharantbine hydroehiorid (CH) and apoptosis induction on a human nasopharyngeal carcinoma cell line SUNE-1. METHODS: MTT method was used to evaluate the growth inhibition effects. Flow eytometry and electron microscopes were used to study the effect of the change of cell cycle and the apoptosis of SUNE 1 cells respectively, which were treated with CH. RESULTS: MTT showed that the CH inhibited SUNE-1 cells growth. There was obvious dosage-effect and time-effect correlation, P〈0.01. The result of flow cytometry showed that cells in Go/G1 phase were in creased after exposure to CH, the proportions of cells in G0/G1 phase were (71.5 ± 8.7)% and those in control group were (46.9± 10.5)%, P〈0.05. That indicated the SUNE 1 cells were blocked in G0/G1 phase. Morphological apoptosis of SUNE 1 cells could be seen under electron microscope. CONCLUSION: CH inhibits SUNE 1 cells proliferation and induces SUNE-1 cells apoptosis.
