摘要
目的:探讨离子霉素对Jurkat T细胞增殖的抑制作用。方法:以Jurkat T细胞为模型,与5种细胞增殖抑制剂比较,观察离子霉素对Jurkat T细胞集落形成的影响,分析不同浓度离子霉素对Jurkat T细胞增殖的影响;分析离子霉素对JurkatT细胞周期的影响;分析离子霉素对Jurkat T细胞凋亡的作用。本实验结果用统计软件包SPSS10.0进行处理,数据以均数±标准差(x±s)表示,多组间比较采用One-Way ANOVA,两组间比较用非配对Student'st检验。结果:随着离子霉素浓度从0.1mg/L逐渐增至4.0mg/L,JurkatT细胞的增殖逐渐减弱,以4.0mg/L离子霉素的抑制作用最为明显,呈剂量依赖关系,且该浓度下,离子霉素对Jurkat T细胞增殖的抑制作用较PD98059、AG490、GF109203X和Genistein明显。离子霉素可使Jurkat T细胞周期停滞于G0/G1期,阻止其进入S期并可促进JurkatT细胞的凋亡。结论:离子霉素对Jurkat T细胞增殖有明显的抑制作用,可使Jurkat T细胞周期停滞于G0/G1期,阻止其进入S期,且对Jurkat T细胞凋亡有显著促进作用。
Objective: To investigate the inhibitory effect of ionomycin on proliferation of Jurkat T cells. Methods: The effect of different concentrations of ionomycin on the proliferation of Jurkat T cells was estimated by methyl thiazolyl tetrazolium (MTT) assay. The comparison of effect of ionomyein with five kinds of selective inhibitors, including PD98059, LY294002, AG490, GF109203X and Genistein on the proliferation of Jurkat T cells was also estimated by MTT assay. Flow cytometry was used to estimated the effect of ionomycin on the cell cycle and apoptosis of Jurkat T cells with propidium iodide (PI) and Annexin V/PI stain. Results: MTT assay and the colony formation of Jurkat T cells showed that the inhibitory effect of ionomycin on Jurkat T cell proliferation was enhanced, following the concentration arising from 0.1 mg/L to 8.0 mg/L. At the concentration of 4 mg/L, the inhibitory effect was the most significant, appearing a dose-dependent manner. At the same concentration, the inhibitory effect of ionomycin was more significant than other four kinds of inhibitory drugs and similar with the effect of LY294002. Analysis of the cell cycle revealed that the cells were blocked into S and phase as the concentration of ionomycin increased. Ionomycin can promote apoptosis of Jurkat T cells at the concentrations from 0.1 mg/L to 4.0 mg/L. Conclusion: Ionomycin can inhibit the proliferation of Jurkat T cells and block their cell cycles into S phase. Ionomycin can promote apoptosis of Jurkat T cells.
