RAPD variation in sweetpotato cultivars from Hunan

摘要 目的:采用RAPD技术对湖南境内的甘薯品种进行多样性分析,并与境外的部份品种进行比较分类与鉴定研究。方法:从100个10bp的随机引物中筛选出20个扩增效果较好的引物进行扩增。结果:共产生188条带,其中118条为多态性带,占62.8%,平均每个引物扩增出的DNA多态性带数为5.90条,根据扩增结果构建出反映品种多样的UPGMA聚状图,31个品种可以划分为4大类。结论:从分子水平揭示了湖南境内部分甘薯品种的亲缘关系。 Objective: Random amplified polymorphie DNA (RAPD) was used to study the classification and identification in sweetpotato cultivars from Hunan, and compare with the other sweetpotato cultivars from outside. Methods: Twenty 10bp primers selected from one hundred arbitrary primers were applied for amplifying the sweetpotato DNA, Results: Total 188 bands were produced, among which 118 bands (62.8%) were polymorphic. The average numbers of polymorphic DNA amplified by each primer were 5.9, These sweetpotato cultivars can be devidde into four groups by UPGMA cluster base on genetic distances. Conclusion: In this study, the RAPD dataset appeared to be appropriate in resolving the sweetpotato varietal relationship.
