盆栽安祖花叶片愈伤组织诱导及植株再生的影响因素 被引量:5

Factors Affecting Callus Induction and Plant Regeneration from Leaf Explants of Pot Anthurium(Anthurium andraeanum Lind.)

摘要 以近几年新引进的6个安祖花盆栽品种为试材,以刚展开的幼嫩叶片为外植体,研究了基本培养基、植物生长调节剂配比、培养条件等因素对其愈伤组织诱导及不定芽分化、增殖的影响。结果表明:MP和1/2MS培养基比MS、135和N6培养基更适于愈伤组织的诱导,1.0~2.0mg/LBA+0.1~0.2mg/L2,4-D对多数品种的愈伤诱导较为合适;MS+0.5mg/LBA+0.5mg/LNAA和MP+1.0mg/LBA适于参试品种的不定芽分化,N6培养基不利于芽的分化;不定芽的增殖以MS+1.0mg/LBA最为合适。不同品种的愈伤组织诱导能力、不定芽分化能力及其增殖效率差异明显。黑暗培养比光照培养更有利于愈伤组织的诱导。理想的生根培养基和移栽基质分别为1/2MS+0.5mg/LIAA和泥炭土与珍珠岩的混合物,生根率与移栽成活率均可达100%。 Using immediately expanded blades as explants,the effects of different basic media,PGR (plant growth regulator) combinations and illumination conditions on the callus induction and adventitious shoot differentiation and proliferation were investigated in six pot cultivars of Anthurium andraeanum. The results showed that MP and 1/2 MS media were better than the MS,B5 or N6 medium for callus induction. 1.0--2.0 mg/L BA plus 0. 1-0. 2 mg/L 2,4-D were suitable for callus induction of most cultivars investigated. MS+0. 5 mg/L BA+0. 5 mg/L NAA and/or MP+1.0 mg/L BA were proper for shoot differentiation of all studied cultivars, while N6 was unfavorable. MS47 1.0 mg/L BA was the optimum medium for shoot multiplication. The differences of callus induction capabilities,adventitious shoot differentiation and proliferation were obvious among different cultivars. Cultures in darkness were better than that under light for inducing calli. The best media for rooting and transplanting were 1/2 MS+0. 5 mg/L IAA and the mixture of peat and perlite, respectively,with 100 % of the rooting frequency and survival rate after transplantation.
