摘要
目的建立测定东方肉穗草中槲皮素含量的方法。方法采用RP—HPLC法,色谱柱为Shimadzu Shimpack VP C18柱(150mm×46mm,5um),流动相为甲醇-0.1%磷酸(45:55),流速为1.0ml·min^-1,检测波长为365nm,柱温为40℃。结果槲皮素0.025~0.400mg·ml^-1与峰面积呈良好的线性关系,回归方程为:Y=7.74×10^-7X-2.69×10^5(r=0.9998),平均回收率为99.8%,RSD=0.83%(n=5)。结论所建方法快速、简便,重复性和稳定性均好,能准确地测定东方肉穗草中槲皮素的含量。
OBJECTIVE To establish a method for determining the content of quercetin in Sarcopyramis bodinieri var. delicata by HPLC. METHODS The Shimadzu Shimpack VP C18 (150 mm × 46 mm ,5 um ) column with methanol -water- phosphoric acid (45 : 55 ) as the mobile phase was used. The flow rate was 1. 0 ml. min - 1 and the detective wavelength was set at 365 nm. The column temperature was set at 40 ℃. RESULTS The calibration curve was linear in the range of 0.025 - 0.40 mg·ml^-1 , Y = 7.74 × 107 X - 2. 69 × 10^5 ( r = 0. 9998 ). The average recovery was 99.8% with the RSD of 0.83%. CONCLUSION The method is simple, accurate, reproducible and can be used for the quality control of Sarcopyramis bodinieri vat. delicata.
