摘要
目的比较不同种属和不同药用部位鬼针草中没食子酸的含量。方法采用RP—HPLC法,用Kromasil C18柱(250mm×4.6mm,5um),柱温为25℃,流动相为甲醇-0.1%磷酸水溶液(8:92),检测波长为274nm,流速为1ml·min^-1。结果没食子酸进样量0.03~0.30ug与峰面积具有良好的线性关系,回归方程为:Y=8.108×10^4X-3.160×10^4(r=0.9999,n=6),平均回收率为101.2%。三叶鬼针草根、茎、叶和狼把草、白花鬼针草、婆婆针、三叶鬼针草中没食子酸的含量分别为6.16%、6.95%、9.26%、6.99%、8.5%、9.78%、10.3%。结论所用方法简便、准确、重复性好,可作为控制鬼针草属药材质量的方法;从没食子酸含量的角度考虑,以三叶鬼针草的叶人药最好。
OBJECTIVE To determine the content of gallic acid from different medical parts and different species of Herba Bidens. METHODS HPLC method was adopted . The analysis was carried out at 25℃ on a Kromasil C18 column eluted with a mobile phase consisting of a mixture of methanol - 0.1% phosphoric acid ( 8 : 92). The flow rate was 1 ml- min 1 , and the detection wavelength was set at 274 nm. RESULTS The calibration curve was linear within the concentration range of 0.03 - 0.30 ug ( r = 0. 9999 ). The average recovery was 101.2%. The contents' in the root, stem and leaf of Bidens pilosa L. were 6. 16% ,6.95% and 9.26% ,respectively. And the contents of Bidens tripartica Linn. ,Bidens pilosa L. var. ratiata Sch - Bip. ,Bidens bipinata Linn. and Bidens pilosa L. were 6. 99% ,8.50% ,9.78% and 10.30% , respectively. CONCLUSION This method is simple, accurate, and suitable for the determination of gallic acid in Herba Bidens. The content of gallic acid in Bidens pilosa L. and the leaf of Bidens pilosa L. was the highest. Considering to the content of gallic acid, it was suggested that the leaf of Bidens pilosa L. was the best, and Bidens pilosa L. was better than others.
