Determination of α-tocopherol in Hippophae oil by HPLC

摘要 目的建立一种简便的测定沙棘油中α-生育酚含量的方法。方法以无水乙醇提取沙棘油中的α-生育酚,用HPLC测定,固定相为C18(15mm×4.6mm,5um)色谱柱,流动相为甲醇-水(95:5),流速为1.0ml·min^-1,检测波长为290nm。结果α-生育酚与其他醇溶性成分分离良好;其浓度2.4~22.0mg·L^-1与峰面积的线性关系良好(r=0.9999);平均回收率为96.81%。结论与传统皂化法比较,所建方法更为简便、快速和准确。 OBJECTIVE To establish a simple HPLC method for the determination of α - tocopherol in Hippophae oil. METHODS Absolute ethanol was used to extract α - tocopherol from Hippophae oil and the content was determined by HPLC. The separation was performed on a Cls column (15 mm ×4.6 mm,5 um). The mobile phase consisted of methanol -water (95 : 5) with the flow rate of 1.0 ml·min^-1. The detection wavelength was set at 290 nm. RESULTS α -Tocopherol could be well separated from other components soluble in ethanol. The standard curve was linear in the concentration range from 2.4 to 22.0 rag. L- 1 ( r = 0.9999). The mean recovery was 96.81%. CONCLUSION The established method is simpler, quicker and more accurate than the traditional method of alkali saponifing. It is suitable for assaying the content of α - tocopherol in Hippophae oil.
