鲨鱼软骨多糖抑制肿瘤生长的实验研究 被引量:6

Empirical study on inhibiting tumor growth from polysaccharide extracted from shark cartilage

摘要 目的:研究鲨鱼软骨多糖对血管内皮细胞生长、新生血管形成及小鼠实体瘤生长的影响作用。方法:以MTT法测定鲨鱼软骨多糖体外对内皮细胞增殖的影响;用鸡胚绒毛尿囊膜实验测定对血管生成的影响;建立荷瘤小鼠,并观察该提取物对小鼠抑制瘤的影响。结果:鲨鱼软骨多糖能显著抑制血管内皮细胞的生长,MTT法抑制率为(45.615±0.084)%;并可抑制鸡胚绒毛尿囊膜新生血管的形成,PBS对照组血管数目为18.750根,鲨鱼软骨多糖组血管数目为(7.750±1.545)根,差异有统计学意义,P<0.05。鲨鱼软骨多糖能抑制荷Lewis小鼠肿瘤生长,鲨鱼软骨多糖组肿瘤抑制率与生理盐水组相比差异有统计学意义,P<0.01;鲨鱼软骨多糖+环磷酰胺组则明显高于环磷酰胺组,差异有统计学意义,P<0.05。结论:鲨鱼软骨多糖可以抑制内皮细胞生长及新生血管的形成,进而发挥抗肿瘤作用。 OBJECTIVE: To study the effect of polysaecharide extracted from shark cartilage on the vascular endothelial cell's (VEC) growth, neovascular formation and solid tumor's growth of mice. METHODS: The MTT method was used to observe the influence of the polysaccharide on the endothelial hyperplasy in vitro. The influence on angiogenesis was determined with embryonated egg allan tocherion and tumor-bearing mice was constructed, and the influence of the polysaceharide on inhibiting tumor was observed in the mice. RESULTS: The polysaccharide significantly inhibited VEC's growth [inhibitory ratio with MTT: (45. 615±0. 084)%)] and the formation of angiogenezis of embryonated egg (the number of blood vessel in the PBS control group was 18. 750, while in the polysaccharide test group was 7. 750±1. 545, and the difference had a statistical significance, P〈0. 05). The polysaceharide inhibited tumor growth in mice that were bearing lung cancer tumor (the difference between polysaecharide and physiological saline had statistical significance, P〈0.01; the number of the polysaccharide + CTX group was significantly higher than that of the CTX group, the difference had statistical significance, P〈0.05 ). CONCLUSIONS: Polysaccharide extracted from shark cartilage can inhibit the growth of VEC and the formation of neovessels. So it can inhibit the growth of tumor effectively.
