大鼠实验性肠系膜上动脉闭塞性休克时胃窦粘膜D和G细胞免疫组织化学研究 被引量:4

IMMUNOHISTOCHEMICAL STUDY OF D- AND G-CELLS IN THE ANTRAL MUCOSA OF THE RATS DURING SUPERIOR MESENTERIC ARTERY OCCLUSION (SMAO) SHOCK

摘要 本实验用成年雄性Wistar大鼠37只,分为休克组、手术对照组和空白对照组。休克组大鼠在麻醉状态下,复制肠系膜上动脉闭塞性休克,并在小肠缺血1小时和1小时后再恢复小肠血流30分钟、90分钟等3个时间取材。手术对照组只重复休克组的手术操作而不阻断小肠血流,与休克组同时间取材。空白对照组不经任何处理,于麻醉后取材。取各组大鼠胃窦组织,制成4μm的连续切片。用免疫组织化学PAP法,在相邻切片上分别显示生长抑素细胞(D细胞)和胃素细胞(G细胞),并进行D和G细胞计数(个/mm^2粘膜)及计算G/D细胞比值。空白对照组D细胞计数为64.77±3.64,G细胞计数为98.54±6.52((?)±S(?)),G/D细胞比值为1.53±0.21((?)±S)。手术对照组的各项指标与空白对照组无显著差异(P>0.05)。休克组在恢复小肠血浣后30分钟和90分钟时,D细胞计数比相应手术对照组的分别下降了27.9%(P<0.05)和36.4%(P<0.01);G细胞计数则无显著差异(P>0.05);G/D细胞比值分别上升了45.1%(P<0.01)和38.5%(P<0.01)。结果提示,在肠系膜上动脉闭塞性休克过程中,大鼠胃窦粘膜D细胞数量的变化,反映了D细胞的分泌活动增强。G/D细胞比值上升,说明胃窦内分泌发生了变化。 37 Wistar rats were divided into 3 groups: SMAO shock group, operation control group and normal control group. The antral tissues of stomach were prepared, in 4μn thick serial sections, for immunohistochemical staining at definite time interval (0, 30 and 90 minutes after shock) in the shock group. On the adjacent serial sections, Sternberger's PAP method was specifically used to demonstrate the somatostatin immunoreactive cells (D-cells) and the gastrin immunoreaetive cells (G-cells) in order to observe their changes during SMAO shock. D- and G-cells counts (cell/mm^2 mucosa) and G/D cell ratio were calculated. In the normal control group, the D-cell count was 64.78±3.64 and that of G-cells was 98.54±6.52((?)±S(?)) , the G/D cell ratio was 1.53±0.21((?)±S) . There was no significant difference (P>0.05)in D-and G-cells counts and G/D cell ratio between the operation and the normal control groups. But, in the shock group at 30 and 90 minutes, D-cell count was 27.9% (P<0.05) and 36.4% (P<0.01) lower than the relevants of the operation control group, G-cell count showed no marked difference and G/D cell ratio was 45.1% (P<0.01) and 38.5% (P<0.01) higher than the relevants of the operation control group. The results suggest that the declined D-cell count may be related to the increased secretion of these cells during SMAO shock and G/D cell ratio increase may suggest the change of the balance between D-cells and G-cells in the antral mucosa during the shock.
