摘要
通过细胞工程手段,使人早幼粒白血病突变株细胞(HL-60-AR)在体外成熟分化。HL-60-AR白血病细胞在与小鼠外周血中性粒细胞融合所产生的杂交细胞获得了粒细胞的成熟分化标志,同时细胞增殖速度减低、细胞在裸鼠中失去致瘤性。在体外软琼脂培养基中虽有集落形成,但其集落形成率有所降低。
It has been well documented that human promyelocytic leukemia HL-60 cells could be induced to differentiate along myeloid pathway by various compounds. In this paper, differentiation of a human promyelocytic leukemia cell mutant (HL-60-AR)was achieved by cell engineering technique.The hybrid cells (HL-W) produced by the fusion of HL-60-AR cells and mouse neutrophil granulocytes were found to differentiate and have obtained the differentiated markers of mature granulocytes.Meanwhile, the proliferation ability of hybrid HL-W filial cells was decreased and also lost tumor-producing ability in nude mice.Although HL W cells could form colonies in soft agar medium, the ratio of colony-forming of hybrid cells was remarkedly decreasad.It would be reasonable to speculate that mouse neutrophil granulocyte might contain endogenous inducing factor(s).
