摘要
目的:观察布地耐德混悬液雾化吸入对哮喘模型肺组织STAT6的干预作用。方法:BALB/C小鼠被随机分为空白对照组、哮喘模型组、激素干预组,除空白对照组外其他各组小鼠在试验第1天、7天和第14天通过腹腔注射0.2 m l混有40 mg氢氧化铝和10μg OVA的pH 7.4 PBS液致敏,在试验第15天用5%的OVA对小鼠进行雾化激发,连续1周,激素干预组在激发的同时给予布地耐德混悬液(1 mg)雾化吸入1周。最后1次激发后24 h放血杀死小鼠,检测血浆IgE水平和血中嗜酸细胞计数,留取肺组织标本用10%的甲醛固定,通过免疫组化的方法对各组小鼠肺组织STAT6的表达进行检测。结果:布地耐德混悬液雾化吸入明显抑制了哮喘模型肺组织STAT6的表达,阻止了哮喘模型鼠气道过敏性炎症。结论:布地耐德混悬液雾化吸入不但可以减轻哮喘模型气道的过敏性炎症,同时可以通过抑制肺组织STAT6的表达进而抑制过敏性炎症的信号传导系统。
Objective : To observe the preventive effect of budesonide on inflammatory signal transduction system of allergic asthma models. Methods: BALB/C rats were divided into control group, asthma models group and intervention group. Rats in asthma model group and intervention group were injected 0. 2ml allergic solution (40 mg of aluminum hydroxide, 10 p.g of OVA, pH 7. 4) on the first, seventh and fourteenth day, then stimulated by 5% OVA on the fifteenth day for 7 days, while rats in intervention group inhaled budesonide suspension for 7 days. The level of IgE in serum and the amount of acidophils in blood were detected, the expression of STAT6 in lung tissues was detected by immunohistochemical staining. Results: Budesonide inhibited the expression of STAT6 in lung tissues of allergic asthma models, prevented the allergic inflammation of asthma models. Conclusion: Budesonide alleviates the allergic inflammation of asthma models, inhibit the expression of STAT6 in lung tissues and the signal transduction system of allergic asthma models.
