摘要
用BGC-823细胞系经吐温80合并39℃~43℃、20分~100分作用后。观察即时,48小时的SDH组化形态及其相对活性的变化。实验表明:合并作用即时最显著,随作用温度、时间增强胞质内SDH颗粒变细、减少并出现浅粉红色小泡。41℃100分作用后多数细胞颗粒消失、充满小泡,43℃后小泡减少,细胞渐成透明状。活性测定各组协同效应明显、41℃20分超过43℃100分作用效应,48小时后仍有协同效应。
Cells of human stomach cancer cell line BGC-823 were incubated at 39℃ , 41℃ and 43℃, for 20, 60. 100 min respectively combining with Tween-80. SDH (succinate dehydrogenase) of BGC-823 cells was observated by histochemical) method at 0 hour (instant groups) and 48 hours (48 hours groups) after treatment. The relative activity of SDH was measuied quantitatively with microspectro-photometer (Leitz MPV-Ⅱ).We found that SDH positive granules in the instant groups were less than that of only hyperthermia. With temperature rising and time extending, SDH granules became fine, dispersed and diminished. Meanwhile, some small pink bubbles appeared in the cytoplasm of the cells. At 41℃ 100min, SDH granules disappeared and small bubbles filled the cytoplasm inmost cells These small bubbles reduced gradualty at 43℃ and most cells became transparent at 43℃ 100min-In 48 hours grotps, the SDH granules further diminished and the small bubbles also reduced. Quantitative analysis showed that SDH relative activity decreased remarkably in instant groups.The coopeiative effects were obvious. The effects at 41℃ 20 min exceeded that of only heating with 43℃ for 100 min, i.e. reducing about two degrees centigrade.Some cooperative effects were still remained in 48 hours groups but not so remarkable as instant groups.
