头孢曲松钠治疗大肠杆菌所致疾病机制的研究

摘要 目的探讨头孢曲松钠对大肠杆菌(E.coliK1)的作用机制。方法采用对照实验,培养E.coliK1至最佳状态,实验组细菌加入头孢曲松钠100μg,对照组加入生理盐水,孵育1h。通过western-blot方法检测E.coliK1外膜蛋白A的表达变化,软件分析蛋白表达灰度值。结果实验组OmpA与对照组比较,表达量明显下降。结论头孢曲松钠可以诱导E.coliK1的外膜蛋白A表达下降,从而影响E.coliK1外膜的完整性。 Objective To investigate the mechanism of ceftriaxone sodium on the invasion of E.coli K1. Methods E.coli K1 were grown overnight in brain heart infusion broth. 100μg ceftriaxone sodium was added to the conflucence E. coli K1 (experimental group) , and incubation for 1 hour. The 0.9% sodium chloride was used as the control( control group). The alteration of OmpA expression in E. coli K1 was detected by western-blot. The level of OmpA expression was analyzed by BandScan software. Results The expression of OmpA in experimental group was significant lower than that of the control group. Conclusion Ceftriaxone sodium can change the integrity of E. coli K1 outer membrane by decreasing the expression of outer membrane protein OmpA.
