嗅三针对血管性痴呆大鼠学习记忆功能及海马八肽胆囊收缩素含量的影响

摘要 目的:探讨嗅三针对血管性痴呆大鼠学习记忆功能及海马八肽胆囊收缩素含量的影响作用。方法:成年SD雄性大鼠40只,体重300±10g。随机分为4组,每组10只。①正常对照组,②VD模型组,③VD嗅球损毁模型组,④嗅三针组。制作VD大鼠模型和VD并损毁嗅球大鼠模型,通过嗅三针电刺激方法,测试大鼠水迷宫学习记忆能力并测定海马八肽胆囊收缩素含量。结果:定位航行试验显示,平均逃避潜伏期比较,VD模型组明显长于正常对照组(P<0.01);嗅三针组明显短于VD模型组(P<0.01);嗅三针组也明显短于VD嗅球损毁模型组(P<0.01);VD嗅球损毁模型组与VD模型组之差异无显著性意义(P>0.05)。空间探索试验显示,原平台象限跨越相应平台的次数比较,VD模型组明显少于正常对照组(P<0.01);嗅三针组明显多于VD模型组(P<0.01);嗅三针组也明显多于VD嗅球损毁模型组(P<0.01);VD模型组与VD嗅球损毁模型组之差异无显著性意义(P>0.05)。海马八肽胆囊收缩素含量比较,VD模型组低于正常对照组(P<0.05);嗅三针组高于VD模型组及VD嗅球损毁模型组(P<0.05);VD模型组与VD嗅球损毁模型组之差异无显著性意义(P>0.05)。结论:嗅三针能够显著增强血管性痴呆大鼠学习记忆功能、能提高海马八肽胆囊收缩素的含量,其治疗效应的发挥依赖于嗅觉传导通路密切相关。 Objective: To explore the effect of XIU Three Needles on learning - memory abillties, and cholecystokinin - 8 (CCK - 8 ) content of hippocampus in the vascular dementia ( VD ) rats. Methods: The 40 Sprague Dawley rats were randomly divided into control, VD model, VD plus removed olfactory bulb model and XIU Three Needles groups. VD models were established by 4- vessel occlusion. Morris maze tests were conducted for valuating the learning and memory abilities. The content of CCK - 8 of hippocampus were detected.Results: In comparison with control group, the average escape latency prolonged significantly, and the targetplatform crossing times decreases remarkably in VD model group ( P 〈 0.01 ) .In comparison with VD model group, the average escape latency shortened and the targetplatform crossing times increases both remarkably in XIU Three Needles group ( P 〈 0.01) .No significant differences were found between VD model and VD plus removed olfactory bulb model groups in the escape latency shortened and the tar- get - platform crossing times ( P 〉 0.05) . The CCK- 8 content of hippocampus in VD model group was lower than that of control group ( P 〈 0.05) ; THe CCK - 8 content of hippocampus in XIU Three Needles group was higher that that of VD model group ( P 〈 0.05 ) ; No significant differences were found between VD model and VD plus removed olfactory bulb model groups in the CCK - 8 content of hippocampus ( P 〉 0.05) . Conclusion: The effect of XIU Three Needles dramatically increased the learning - memory abilities of VD rats, increased CCK- 8 content of hippocampus in VD rats at the same time.Its therapeutical effects closely related to the olfactory pathway.
