摘要
目的:探讨用胰岛素提高人肺腺癌细胞A549生长代谢水平及其机制。方法:采用MTT比色法,分析胰岛素对肺腺癌A549细胞代谢的影响,用流式细胞仪作细胞周期分析,用Western Blot印迹方法检测细胞周期蛋白CyclinA和CyclinD1的表达情况。结果:胰岛素(2.0~32.0mU/mL)可促进人肺腺癌细胞A549的增殖(P<0.05),细胞周期结果显示胰岛素(8mU/mL)可使S期细胞增多。Western Blot印迹方法检测结果显示胰岛素(8mU/mL)后CyclinD1的表达明显增强,CyclinA的表达变化不明显。结论:胰岛素能促进人肺腺癌细胞株A549增殖,并且其增殖作用是可逆的。胰岛素主要作用于人肺腺癌细胞株A549细胞的S期。CyclinD1的表达增强在胰岛素促增殖机制中起着重要作用。
Objective: To investigate the effect and mechanism of insulin on human lung adenocarcinoma cell strain A549. Methods : Effect of insulin on the metabolism of human lung adenocarcinoma cell strain 3.549 was analyzed by MTT. Cell cycle was analyzed by flow cytometry (FCM). Expression of cyclins A and D1 was detected by Western blot. Results: Insulin at the dose of 2.0 - 32.0mU/mL promoted the proliferation of human lung adenocarcinoma cell strain A549 ( P 〈 0.05). Cell cycle analysis showed that insulin at the dose of 8mU/mL increased the number of human lung adenocarcinoma cells at S phase ( P 〈 0.01 ). Western blot displayed that the expression of cyclin D1 was significandy increased, while that of cyclin A remained unchanged after treatment with insulin at the dose of 8mU/mL. Conclusion: Insulin promotes the proliferation of human lung adenocarcinoma cell strain A549, and acts mainly on human lung adenocarcinoma cell strain 3.549 at S phase. Increased expression of cyclin D1 plays an important role in the proliferation of human lung adenocarcinoma cell strain A549.
