神经生长因子对大鼠脑缺血再灌注后HIF-1α及HSP70表达的影响

Effect of nerve growth factor on expression of hypoxia-inducible factor 1 in rat after cerebral ischemical reperfusion

摘要 采用大鼠大脑中动脉(MCAO)模型,肌肉内注射神经生长因子(NGF),经过免疫组化和HE染色检测Sprague-Dawley(SD)大鼠脑缺血再灌注后HIF-1α(缺血诱导因子)及HSP70(热休克蛋白70)表达的变化及神经生长因子的影响。结果表明,肌肉注射神经生长因子对脑缺血再灌注有明显的保护作用,HIF-1α及HSP70表达增加可能是其机制之一。 Applying middle cerebral artery occlusion model of rat and injecting nerve growth factor in muscle and staining with immunohistochemical method and HE method to observe the change of expression of hypoxia - inducible factorl ( HIF -1α) and heat shock broken 70(HSP70) and the effect of nerve growth factor on it after focal cerebral ischemical reperfusion injury in Sprague - Dawley (SD) rat. The expression of heat shock broken 70 increased after cerebral ischemical, reperfusion injury and peaked at 24th hour, and reduced by then. Compared with the group of ischemical reperfusion, the expression of heat shock broken 70 increased after injection of nerve growth factor in rats, (P〈0. 01 ). The results implied that injection nerve growth factor in muscle was protective effect clearly on cerebral ischemical reperfusion injury. The increasing of expression of hypoxia - inducible factor and heat shock broken 70 is the principle probably.
