摘要
目的:探讨热瘀散治疗胃肠湿热型溃疡性结肠炎(ulcerative colitis,UC)的作用机制。方法:雄性Wistar大鼠50只,随机分为正常对照组、模型组、柳氮磺胺吡啶(salicylazosulfapyri-dine,SASP)组、热瘀散低剂量组、热瘀散高剂量组。采用复合因素(高脂、高糖饮食加湿热环境)复制胃肠湿热证模型,再用2,4,6-三硝基苯磺酸(trinitro-benzene-sulfonic acid,TNBS)与500 ml/L乙醇复合溶液复制溃疡性结肠炎模型。模型复制后各组大鼠给予相应药物治疗,连续15 d,第16天处死动物,观察大鼠结肠组织病理变化,检测血清白介素(interleukin,IL)-8和IL-10的含量。结果:模型组大鼠结肠组织大体评分、病理组织学变化表明模型复制成功,热瘀散明显改善结肠组织的病理损伤,热瘀散高、低剂量及SASP均可降低血清IL-8水平,提高血清IL-10水平。结论:热瘀散通过抑制促炎细胞因子,促进抗炎细胞因子生成,发挥对UC的治疗作用。
Objective:To explore the mechanism of Reyusan, a traditional Chinese medicine for dampness-heat syndrome of stomach and intestine , in treating ulcerative colitis ( UC ) . Methods : Fifty healthy , male Wistar rats were averagely randomized into normal control group, model group, salicylazosulfapyridine (SASP) group, low (lose of Reyusan group and high (lose of Reyusan group. The dampness-heal syndrome of stomach and intestine was induced by composite factors including high fat, high glucose diet and humid heat surroundings. The UC model was induced by the combination of trinitro-benzene-sulfonic acid (TNBS) and alcohol 500 ml/L. After the replication of animal model, rats in each group were daily treated with corresponding drugs during 15 days. Sixteen days later, the rats were sacrificed, the pathological changes of colon tissue were observed, and the levels of interleukin-8 (IL-8) and interleukin-10 (IL-10) were assayed. Results: The general scoring and pathological changes showed that the UC model was successfully induced. Reyusan markedly improved the pathological changes. The low and high doses of Reyusan and SASP all could decrease the level of II-8, increase the level of IL-10. Conclusion: Reyusan achieves a therapeutic effect by inhibiting the production of proinflammatory cytokines, improving the production of antiinflammatory cytokines.
