摘要
研究在压力负荷下阿托伐他汀对钙调神经磷酸酶介导的心肌肥大的影响。选用SD大鼠随机分为3组:假手术组(n=10)、单纯模型组(n=10)和阿托伐他汀组(n=10)。大鼠通过腹主动脉缩窄建立压力超负荷模型,8周后测定左室重量指数,B超检测左室形态结构,Western blot检测心肌CaN蛋白表达,RT-PCR法检测心肌CaN mRNA水平。结果①单纯模型组和阿托伐他汀组心肌肥厚指数明显高于假手术组,阿托伐他汀组心肌肥厚指数明显低于单纯模型组(P<0.05)。②单纯模型组和阿托伐他汀组心肌CaN蛋白及CaN mRNS表达水平高于假手术组(P<0.05),阿托伐他汀组低于单纯模型组(P<0.05)。说明阿托伐他汀可能参与干预钙调神经磷酸酶介导的通路,从而抑制心肌肥厚的形成。
To investigate the effects of atorvastatin on cardiac hypertrophy mediated by calcineurin in rats with pressure-overload, Sprague-Dawley rats were randomly allocated into three groups: The sham-operated groups (n = 10), model groups(n = 10) and atorvastatin-treated groups( n = 10). The model of pressure overload was established by abdominal arota constriction. At the end of the treatment period, the left ventrieular weight/body weight ratio , left ventrieular mass, wall thickness( by echocardiography) , myocardial content of CaN and CaNmRNA( by westernblot and RT-PCR) were assessed . It is resulted that ① The left ventricular weight/body weight ratio of model group, as well as atorvastatin-treated group , was significantly higher than sham-operated group ( P 〈 0.05 ) ; compared with model group, the ratio was lower in atorvastatin-treated group ( P 〈 0.05 ). ②observed a higher level of CaN and CaNmRNA expression in model group, as well as atorvastatin-treated group, as compared to shamoperated group( P 〈 0.05 ) ; significant higher level of CaN and CaNmRNA expression was observed in model group compared to atorvastatin-treated group ( P 〈 0.05 ). The present data suggests that atorvastatin may inhibit calcineurin-mediated hypertrophy in rats with pressure-overload.
