摘要 目的:研究脾虚模型脑内IL-10水平与基因表达变化及益气扶正中药的干预作用。方法:采用苦降泻下、饮食失节加劳倦过度法建立脾虚大鼠模型,免疫组化法与原位杂交法观察下丘脑腹侧核、海马CA1区IL-10水平与IL-10NmRNA表达变化及四君子汤、玉屏风散干预作用。结果:模型组IL-10水平与IL-10mRNA表达在下丘脑腹侧核、海马CA1区明显增加;治疗组上述脑区的IL-10水平与IL-10mRNA表达明显降低。结论:四君子汤、玉屏风散可通过调控IL-10水平及基因表达而影响机体免疫功能。 Objective:To observe the level changes of LI - 10 and gene expression in the brain of rats with spleen defi- ciency, influence of replenishing Qi to invigorate the spleen and strengthening the body resistance. Methods:To establish the rat model of spleen - deficiency by the methods of purgation with bitter and cold descend, diet to excess, hunger and be overstrain. The method of Immunohistochemical and in situ hybridization were used to detects LI - 10 level and gene expression in the hypothalamus ventral kernel, the hippocampal CA 1 region. Results:The immunity positive reactant of LI - 10 in above brain zones increased obviously in the model group, while deceased obviously in the treatment group. LI - 10 mRNA expression in the hypothalamus ventral kernel and the hippocampi CA 1 area increased obviously in the model group, while deceased obviously in the treatment group. Conclusion:LI - 10 (with the effect of inhibit immunity abilities) level and gene expression in the brain of spleen - deficiency rat model has changed. Sijunzi Decoction and Yupingfeng Decoction can influence the immunity abilities by regulating LI- 10 level and gene expression.
