丹参干预骨髓间充质干细胞移植治疗急性心肌梗死的实验研究 被引量:9

摘要 目的:对比研究经静脉移植骨髓间充质干细胞(MSCs)及骨髓间充质干细胞联合中药移植对心肌梗死兔的血清P-selection(P-选择素)的影响,推测干细胞移植的安全性,以及药物干预骨髓间充质干细胞移植对心功能的影响。方法:取兔骨髓间充质干细胞进行体外培养、增殖和标记,同时将已建立心肌梗死模型的日本大耳白兔随机分为4组,模型对照组(B组),静脉0天注射干细胞组(C组)、静脉0天注射丹参组(D组)、静脉0天注射丹参联合干细胞组(E组),开胸结扎冠状动脉前降支的方法建立急性心肌梗死模型,造膜即刻经静脉注射已经分离、培养、扩增、标记后的MSCs悬液,静脉注射中药组经静脉注射丹参注射液。移植后24h检测外周血血清肌酸激酶同功酶(CK-MB),移植后24h及5周检测外周血中P-selection含量,探讨干预因素对内皮功能的影响。移植后4周测定心功能。结果:超声检查显示,与A组相比,B、C、D各组心功能明显降低,差异有统计学意义;与B组相比,E组心功能明显改善,差异有统计学意义,C、D组虽有心功能改善,但不具统计学意义,C组与D组间无统计学差异。血清P-选择素检测示:与B组比较,C、D、E各组均有明显的降低,差异有统计学意义;C、D、E组间无统计学差异。结论:中药丹参可促进MSCs移植治疗心肌梗死的作用,可有效降低P-选择素的表达,有效的改善心肌梗死兔的心功能。 Objective: To compare intravenous transplantation of bone marrow mesenchymal stem cells (MSCs) and bone marrow mesenchymal stem cells transplantation combined with traditional Chinese medicine for myocardial infarction in rabbit serum P - selection ( P - selectin) the impact that stem cell transplantation security and drug intervention of bone marrow mesenchymal stem cells transplantation on cardiac function. Methods : Rabbit bone marrow - derived mesen- chymal stem cells cultured in vitro, proliferation and tags, at the same time has been established myocardial infarction model of the Japanese white rabbits were randomly divided into 4 group,model control group ( B group) ,0 days of intravenous injection of stem cell group ( C group) ,0 days of intravenous injection of Danshen group ( D group) ,intravenous injection of 0 days Salviae joint stem cell group (E group) ,thoracotomy anterior descending coronary artery ligation method of the establishment of acute myocardial infarction model, building membrane instantly by intravenous injection has been i- solated and cultured, amplified,marking MSCs after the suspension, intravenous medicine group by intravenous injection of Danshen Injection. 24 hours after transplantation of peripheral blood serum creatine kinase isoenzyme (CK- MB), and 24 hours after transplantation of peripheral blood 5 weeks of P - selection content ,explore the intervention factors on endothelial function. 4 weeks post - transplant cardiac function measured. Results: The ultrasound examination showed that compared with the A group, B, C, D in each group significantly decreased cardiac function, the difference has statistical significance;and B group, E group significandy cardiac function improvement, the difference has statistical significance, C, D Group although the cardiac function improved, but not statistically significant, C group and D group I statistical difference. Serum P- selectin detection show: with the B group, C, D, E in each group were significantly lower,the difference has statistical significance;C, D,E group no statistical difference. Conclusion: The traditional Chinese medicine Danshen MSCs transplantation can promote the role of the treatment of myocardial infarction,which can effectively reduce the P- selectin expression, an effective myocardial infarction to improve cardiac function of rabbits.
