摘要
目的通过体外实验,观察不同体积分数一氟二氯乙烷(F-141b)对中国仓鼠肺成纤维细胞(V79细胞)的毒性作用。方法用不同体积分数(10、20、30、40、50和60 ml/L)且经抽滤的一氟二氯乙烷对V79细胞进行染毒,分别培养不同时间(3、6、12、24和48 h)后,于显微镜下计数存活细胞数,计算存活率和IC50。同时设阴性对照组。结果在任一培养时间段,细胞存活率均随染毒剂量增加而降低,短时间组(3 h、6 h)和长时间组(24 h、48 h)组间比较差别具统计学意义(P<0.05);染毒剂量相同,细胞存活率随时间增加而降低,各染毒剂量组间差别显著(P<0.05),其中3 h组、12 h组和48 h组相关具有显著的统计学意义(P<0.05)。存在明显的剂量―效应和时间―效应关系。24 h染毒IC50体积分数为32.37 ml/l。结论在本实验条件下,一氟二氯乙烷具有显著的细胞毒性作用,并存在剂量和时间效应。
Objective To explore the cytotoxicity of F-141b in V79 Chinese hamster cells in vitro. Methods V79 cells suspension were exposed in different concentration (0,10,20,30,40,50 and 60 ml/l) F-141b agent and incubated at 37℃ for different time(3,6,12,24 and 48 h). The number of living cells was counted under microscope. The survival rate and IC50 was observed. Results In any period of definite time, the survival rate of V79 cell decreased gradually with increasing concentration of F-141b. On the other hand, at the same concentration of F-141b, the longer the incubation was, the less the survival rate of V79 cell was. There was clear discrepancy between short incubations(3 h,6 h) and long incubations (24 h,48h) (P〈0.05). And there was also clear discrepancy among different concentration groups(P〈0.05). Otherwise there was statistical significance among co-linearity in 3 h, 12 h,48 h groups. IC50 in the 24 h group was 32.37ml/L. Conclusion With this experiment condition, F-141b could induce cytotoxicity and is in a close-dependent and time-dependent manner.
