摘要
研究发现水飞蓟种仁中存在凝集素(SML)。提取SML时,种仁粉与TrisHCl(pH72)缓冲液的抽提比例为1∶5时较为合适,此时的滴度为24,发生凝集反应的蛋白质浓度为387μg/ml。水飞蓟凝集素的最适pH为6~8。其热稳定性较强,能耐60℃温度处理。其专一性糖为D半乳糖。经硫酸铵分级沉淀。
Lectin was found in Smarianum kernel, appropriate rate of kernel powder and extracting buffer TrisHCl(pH 72) was 1∶5(W/V),under this condition, the titre of SML is 24,and protein concentration is 387 μg/ml The lectin is relatively stable to heat and tolerant to temperature 60℃ The hemagglutinating activity of SML is specially inhibited by Dgalactose Fractional precipitation with (NH4)2SO4 indicated that the component of SML with strongest hemagglutinating activity is 020~040 stad ppt
