Study on the Optimization of Extracting Technology of Chlorogenic Acid from Eucommiae Leaf Powder

摘要 [目的]优选杜仲叶粉中绿原酸的最佳提取方法和提取工艺,为工业化生产提供参考。[方法]用酶解和超声相结合的方法,在单因素试验的基础上,采用四因素三水平正交设计法对提取工艺条件进行优选。[结果]单因素试验结果得出,在溶剂pH值为4.5,酶加入量为0.6%,酶解温度为40℃,料液比为1∶10,超声时间为30 min,绿原酸提取得率最大。在此基础上进行的正交试验结果显示,绿原酸提取的最佳工艺条件为:溶剂pH值为5.0,酶解温度为40℃,料液比为1∶10,超声时间为30 min,在此最佳条件下绿原酸提取得率可达3.05%。从与酶法和超声波法的比较试验得出,绿原酸提取得率比酶解高出5.54%,比超声波提取高出16.68%。[结论]采用酶解与超声波结合的方法,先让酶作用后,有利于超声波的进一步作用,促进绿原酸的释放,提取效果远远超过酶法和超声波法。 [ Objective] The method and technology of the chlorogenic acid extracted from eucommiae powder were optimized for the reference for the development of the industry. [Method] The optimal technology was experimented with the combination of uhrasound enzymolysis method through the orthogonal test of 4 factors at 3 levels based on the experiment in single factor. [Results] The result from the experiment in the single factor was the maximum production of chlorogenic acid extracted from eucommiae powder was under the following conditions: the pH value of solvent, 4.5; the enzyme amount, 0.6% ; the enzymolysis temperature, 40 ℃ ; the ratio of ethanol to folium eucommiae, 1 : 10 and the extraction time with ultrasound, 30 minutes. The resuit from orthogonal test indicated that under the optimal extraction technology including the pH value of solvent, 5 ; the enzymolysis temperature, 40 ℃ ; the ratio of ethanol to folium eucommiae, 1 : 10 and the extraction time with ultrasound, 30 minutes. The output of chlorogenic acid was as high as 3.05%. Compared the method of ultrasound and e nzyrnolysis, the output of chlorogenic acid extracted from eucommiae powder was 5.54% higher than that with the enzymolysis method and 16.68 % higher than that with the ultrasound method. [ Conclusion ] The combination of the ultrasound and enzymolysis method was beneficial to the release of chlorogenic acid because the eucommiae powder was firstly reacted by enzymolysis and then by the ultrasound so that the combination method was much better than single way.
