摘要
[目的]为吸附树脂法分离纯化苹果原花青素奠定一定的基础。[方法]研究通过大孔吸附树脂纯化苹果中原花青素柱层析的最佳条件。[结果]D3520树脂分离原花青素的效果较好。随着乙醇浓度的增加,洗脱的效果趋好,为了得到高的洗脱率,选用浓度70%的乙醇作为洗脱剂;当上样液pH值为7.0时,吸附效果较好;流速太小时,解吸太慢,解吸时间很长,造成单位体积解吸液中的原花青素含量低,可选择2~6ml/min的速度进行洗脱。正交试验结果表明,洗脱液为浓度70%乙醇,上样液pH值为8,洗脱液流速3ml/min为D3520树脂的最佳分离条件,所得产物的苹果原花青素含量和得率都较高。[结论]确定出了性能较佳的D3520树脂,并得到了其最优的吸附、解吸试验条件。
[Objective] The basis of the isolation and purification of proanthocyanidins from apple with macroporous resin was laid through the experiment. [Method] The optimum conditions for proanthocyanidins isolated from apple with the macroporous rosin were studied. [Results] The macroporous rosin - - D3520 was better material for the isolation of proanthecyanidins. With the incroment of ethanol concentration, the elution effect was getting better and better. In order to have a high elution rate, the ethanol with the concentration of 70% could be used as eluant. When the pH value of liquid sample was 7.0, the adsorption effect was good. The low content of Proanthocyanidin could be caused while the flowing speed and resolving was too slow and it was best for the flowing speed to be 2- 6 ml/min. The orthogonal test results showed that the eluate of 70% ethanol, pH value of sample solution of 8, the flowing rote of 3 ml/min was the host isolation condition with high content and yield of proanthocyanidinsa. [Conclusion] The performance of the resin - - D3520 was identified and the optimal adsorption, desorption experimental conditions were obtained.
