摘要
目的:探讨survivin基因真核表达载体的构建,研究它在新疆哈萨克族食管癌中的作用。方法:采用PCR方法从新疆哈萨克族食管癌的cDNA表达阳性的标本中扩增出survivin基因,经双酶切后、与真核表达载体pEGFP-N1相连后转入DH5α。结果:DNA测序证明获得了包含ORF全长的survivin基因,并成功克隆入真核表达载体pEGFP-N1中。结论:构建真核表达载体成功,为进一步探讨survivin基因在哈萨克族食管癌中的作用奠定了基础。
Objective: Investigate the construction of survivin in Kazakh's esophageal cancer Eukaryotic Expression Vector, for further study on Kazak's esophageal cancer. Methods: We cloned survivin gene with PCR from Kazak's esophageal cancer in cDNA expressed positiveness, After restrietive endonuelease enzyme EcoR I and BamH I cut, survivin gene link with Eukaryotic Expression Vector pEGFP-N1 and transfer into DH5a. Results: DNA sequence which included ORF was gained, and succusful linked with Eukaryotic Expression Vector pEGFP-N1. Conclusion: We sucussful construct the Eukaryotic Expression Vector, it's basic for further studying survivin gene effect on Kazakh's esophageal cancer effection.
