感染猪囊尾蚴的家猪组织中不同时间IL-6、IL-8、TNF-α和sIL-2R含量变化 被引量:8

摘要 目的研究实验家猪感染猪囊尾蚴不同时间组织中细胞因子IL-6、IL-8、TNF-α和sIL-2R含量变化,从细胞因子角度探讨家猪抗猪带绦虫免疫应答的机制。方法用猪带绦虫虫卵直接灌胃20d龄健康乳猪4头,并以健康乳猪4头作对照。于感染后第40、60、80和120 d取实验组猪肝脏、骨骼肌、心肌、舌肌及脑组织中囊尾蚴寄生处组织200 mg匀浆,用ELISA法测定匀浆上清液中细胞因子IL-6、IL-8、TNF-α和sIL-2R的含量。结果1.肝脏病变组织中4种细胞因子的含量在40d时均明显高于同期对照组(P<0.01);随感染时间延长,4种细胞因子的含量逐渐降低,至80d时与同期对照组差别缩小(P<0.05),120d时肝脏已无囊尾蚴寄生;对照组肝脏组织中4种细胞因子的含量在不同时间始终保持基本稳定。2.肌肉和脑部份病变组织中4种细胞因子的含量在40d时也明显高于同期对照组(P<0.01);随感染时间延长至80d时与同期对照组差别变小(P<0.05),120d时与同期对照组差别无显著性(P>0.05)。3.对照组家猪肝脏中4种细胞因子的含量绝对值高于肌肉和脑组织(P<0.01);实验组肝组织在40d时各种细胞因子的增量更明显高于肌肉和脑组织中的增量(P<0.01)。结论家猪感染猪带绦虫早期,IL-6、IL-8和TNF-α作为前炎性反应因子在家猪肝脏的高水平表达诱导肝脏发生严重的炎症反应,限制和杀灭肝内的早期囊尾蚴,而肌肉和脑组织中这些细胞因子表达稍低,炎症反应相对较轻,因而囊尾蚴易于在这些部位存活。这一点可以从感染后期肌肉、脑组织中的囊尾蚴数量远多于肝脏而得到证实。 To measure the levels of IL-6, IL-8, TNF-α and sIL-2 receptor (sIL-2R) in tissues of pigs infected with cysticercus cellulosae of Taenia sodium in order to explore the immune mechanism of this infection from standpoint of cytokines and their receptors, four pigs of 20 days old were infected with a dose of 80 000 eggs of T. sodium and the levels of IL-6, IL-8, TNF-αand sIL-2R in the infected tissues of pigs were measured on 40^th ,60^th ,80^th and 120^th day after infection by ELISA assay. It was found that the levels of these cytokines and receptor in liver, skeleton muscle, myocardium, muscles of tongue and brain during the experimental periods in the control group of animal were quite stable, while in the experimental group, those in the liver tissues were obviously different in comparison with those of the the control group 40 days after infection (P〈0.01) ; little difference existed in 80^th day, (P〈.05),and no living cystieercus cellulosae could be detected in liver tissues in 120^th day. Similar condition on the levels of these cytokines and receptor in muscle and brain could be demonstrated. In addition, their absolute values of these substances in Ever tissues of normal controls were higher than those in muscle and brain (P〈0.01), and on 40^th day after infection, their increasing amounts were significantly higher than those in muscle and brain (P〈0.01). From these observation, it is evident that in the early stage of cysticercus cellulosae infection, the high expression of IL-6, IL-8 and TNF-α in liver may induce a intensive inflammatory responses which may limit the growth or even kill the unmature cysti-circus cellulosae in liver, while in muscle and brain, the levels of these cytokines show little difference with those of normal controls and induce slight degree of inflammatory responses, growth of eysticereus cellulosae in these sites.
