Activity Assays for Stress activated Protein Kinase and p38 Protein Kinase

应激活化蛋白激酶（ＳＡＰＫ）和Ｐ３２是丝裂素活化蛋白激酶家系（ＭＡＰＫｓ）的主要成员。与应激和某些炎性细胞因子引起的细胞反应有关，参与心血管疾病、细胞凋亡与机体的应激反应。本文主要介绍用免疫沉淀法提取ＳＡＰＫ和ｐ３８，据ＳＡＰＫ和ｐ３８分别可以磷酸化ｃＪｕｎ和ＡＴＦ２的原理，采用聚丙烯酰胺电泳（ＳＤＳＰＡＧＥ）方法，分别测定ＳＡＰＫ和ｐ３８将３２Ｐ参入其特异性底物的量，反映其活性。

Stress activated protein kinase(SAPK)and p38 protein kinase are the main members of Mitogen activated proteins(MAPKs).They are involved in the cellular reation induced by stresses and inflammation cellular factors,and are associated with cardiovascular diseases,appoptosis,and body's stress reactions.In this paper,the assay methods of the two protein kinases are described.By extracting the two kinases with immunoprecipitation,the activities of SAPK and p38 protein kinase are determined by detecting the 32 P incorporation into their specific substates c Jun(SAPK)and ATF 2 (p38).These methods me asures the activities of SAPK and p38 well.
