摘要
目的:克服亲和层析纯化抗人绒毛膜促性腺激素抗体(抗hCG抗体)IgG中载体与配体联接时不良的空间效应,以提高亲和层析的效率。方法:用6-氨基己酸为手臂通过溴化氰连接到Sepharose4B上形成一种新的层析介质,然后将hCG与之偶联用以纯化抗hCG的抗体IgG。结果:用该种亲和层析介质纯化的抗hCG抗体IgG回收率及免疫滴度均高于无手臂的亲和层析介质(活性回收率平均增加22.5%,比活性平均增加55.6%)。结论:用接手臂层析介质纯化抗hCG抗体IgG是一种比较好的方法。
Objective: To overcome the bad spatial effect in connecting carrier and ligand in affinity chromatography and to improve the efficiency of the latter. Methods: The sepharose 4B attached with 6 aminocaproic acid as spacer arms through BrCN activation was used as a new chromatographic medium, with which hCG was coupled to form an affinity chromatographic medium to purify anti hCG IgG. Results: Both the recovery rate(%) and immunological titers of anti hCG IgG purified with spacer arm sepharose 4B were increased as compared with those obtained when sepharose 4B without spacer arms was used (the activity recovery rate and specific activity increased by 22.5% and 55.6% in average respectively). Conclusion: These results show that the purification of anti hCG IgG with spacer arms affinity chromatographic medium is satisfactory.
