摘要
目的:优化大肠杆菌XL1-blue菌株的电穿孔转化条件。方法:采用不同的条件组合转化大肠杆菌XL1-blue菌株,对转化条件进行优化。结果与讨论:用电穿孔方案转化大肠杆菌XL1-blue菌株,可得到明显高于其他转化方法的转化效率。外加电场强度和作用时间是决定转化效率的关键因素,其间存在一定的互补关系,转化效率和外源基因浓度之间呈线性关系,并与细菌浓度密切相关。
Objective: To optimize an electroporation protocol that yields highly efficient gene transfer in Escherichia coli strain XL1 blue. Methods: A series of affecting factors were tested to find out an electroporation protocol for E.coli strain XL1 blue. Results and Conclusion: We describe and optimize an electroporation protocol that yields highly efficient gene transfer in E.coli strain XL1 blue . Voltage gradient and time constant play central roles in this procedure. Transfection efficiency can be linearly elevated by increasing the amount of DNA, and is influenced by the factors such as temperature and density of host cells.
