摘要
采用pH=8.0低离子强度不连续系统聚丙烯酰胺凝胶电泳方法,分析鸡形目4种鸟(花尾榛鸡、红腹锦鸡、环颈雉和家鸡),6种不同组织乳酸脱氢酶(LDH)同工酶。实验表明:这种凝胶系统电泳方法适合鸟类LDH同工酶的分离,分辨率高、效果好、重复性一致。经热稳定性实验和低浓度尿素抑制实验,这4种鸟肝LDH同工酶对热稳定性和尿素抑制不敏感性,其顺序为LDH1>LDH2>LDH3>LDH4>LDH5。根据对LDH同工酶A、B亚基相对迁移率大小比较,表明松鸡科与雉科之间存在差异。并讨论雉科3个种之间亲缘关系,反映出雉属和锦鸡属关系密切,与家鸡亲缘关系较远。
Low ion density discontinuous polyacrylamide electrophoresis (pH=8 0) of LDH from different tissues of four species (Tetrastes bonasia, Chrysolophus pictus, Phasianus colchicus, Gallusgallus domesticus) in Galliformes showed that this electrophoresis system was suitable for analysing avian LDH. Thermostability to heat and nonsusceptibility to urea inhibition of low-density urea of the four liver LDH s were in such order as LDH 1>LDH 2>LDH 3>LDH 4>LDH 5. And it appears, by comparing relative movable ratio of A and B subunits of LDH s, that there exists difference between Phasianidae and Tetraonidae and in Phasianidae Phasianus is closer to Chrysolophus than to Gallus.
