重组人造血生长因子对抗白血病药物作用的影响

摘要 采用流式细胞仪及INT细胞毒检测,研究造血生长因子(rhHGF)对抗白血病药物细胞毒性的影响,并探讨HGF与化疗药物联合应用的作用机理。研究结果表明:3种抗白血病药物柔红霉素(DNR)、三尖杉酯碱(H)、足叶乙甙(VP16)对白血病细胞敏感性存在较大差异;GMCSF、IL3均能增加VP16、DNR对白血病细胞的杀伤作用;GMCSF能提高H的细胞毒性作用,而对正常骨髓细胞。 To explore whether rhHGF and chemotherpeutic drugs combined treatment is helpful to killing more AML cells,an in vitro study on the combined effects of G CSF, GM CSF or IL 3 plus the chemotherapeutic drugs(DNR VP 16 or H)on the proliferation and cytotoxicity of blasts was carried out in AML patient and in normal bone marrow mononuclear cells(NBMMC).NBMMC from 11 healthy volunteers and AML blasts from 23 patients were incubated for 48h with HGF plus antileukemic drug.The drug sensitivity showed marked variation from case to case. The results showed enhancement of the cytotoxicity of DNR,VP 16 by GM CSF or IL 3. The results suggest significant enhancement of the H cytotoxicity for the AML samples by GM CSF.However,none of the HGF significantly effects the drug cytotoxicity of NBMMC.
