erbB-2表达抑制与表皮生长因子刺激对信号转导与转录激活分子及细胞周期蛋白D1的影响 被引量:5

摘要 目的:观察胃癌细胞内原癌基因erbB2表达降低前后信号转导与转录激活分子(STAT1)和细胞周期蛋白D1(cyclinD1)对表皮生长因子(EGF)刺激的反应性。方法:采用Westernbloting检测erbB2表达下调前后及EGF刺激前后STAT1和cyclinD1水平。结果:erbB2表达抑制对STAT1的EGF反应性无显著影响;erbB2表达下调后cyclinD1的EGF可诱导性显著降低。结论:erbB2表达降低可抑制EGF对cyclinD1的诱导效应。 Objective: To investigate the protein level of STAT1 and cyclin D1 following EGF stimulation before and after the repression of erbB-2 gene. Methods: Western blotting analysis of STAT1 and cyclin D1 following EGF stimulation before and after the decrease on erbB-2 expression. Results: Alteration on erbB-2 expression has little effect on the level of STAT1 upon EGF stimulation; induction of cyclin D1 upon EGF stimulation significantly decreased after the repression of erbB2. Conclusion: Decrease of erbB2 expression can suppress the EGFinduction of cyclin D1.
