摘要
目的:培养人牙囊细胞(DFCs),观察陶瓷化骨(CBB)对人牙囊细胞生长和分化的影响。方法:体外分离胚胎期人DFCs,将第三代细胞以CBB为载体立体培养,以平面接种为对照组,用矿化液持续诱导,利用光镜观察细胞生长状态、免疫组织化学观察细胞矿化基质牙骨质附着蛋白(CAP,)及骨涎蛋白抗体(BSP)分泌、Vokassa染色观察矿化情况,了解CBB对DFCs生物学行为的影响。结果:体外连续诱导28d,实验组光镜下可见DFCs与CBB相容性好,材料周边细胞生长密集,并有向BBC趋化迁移现象,附近细胞聚集,形成Vokassa染色阳性的矿化结节,免疫组织化学染色BSP,CAP阳性着色,部分细胞强阳性着色。对照组细胞复层生长,未形成矿化结节,免疫组织化学染色BSP,CAP阴性着色。结论:体外培养条件下,CBB可诱导人DFCs向成骨和成牙骨质细胞分化。
Objective: To investigate the Effect of ceramic bovine bone on behave of purified human dental follicle cells. Method:Human dental follicle cells was cultured in vitro.The subsequent cell passaging were conducted in conditioned medium containing dexamethasone,beta-sodium glycerophosphate and ascorbid acid.with the human dental follicle cells in curture then divided into BBC (in which BBC was added in T-flask)and Hdfc group (without BBC). Migration was observed under inverted phase microscope and differentiation was observed with Von Kasso stain and. Result: BBC improved the migration and differentiation of cells cultured in vitro, showing no difference in proliferation between two grops. Conclusion: BBC can improved the migration and differentiation of human dental follicle cell.
