摘要 采用非同位素聚合酶链反应—单链构象多态性技术,对30例横纹肌肉瘤组织p53基因第5~第8外显了进行检测。结果发现30例横纹肌内瘤中,7例有异常移动电泳带(23.3%)。其中2例同时有第7、第8外显子点突变。此法的结果受多种因素的影响,提示要提高阳性率和正确的可重复的结果,必须有严格的实验环境和条件。 Non isotopic polymerase chain reaction (PCR) and single stranded conformation polymorphism (SSCP) method was used to detect point mutation of p53 gene in 30 cases paraffin embedded rhabdomyosarcomas (RMS). The abnormal shifting of the singlestranded DNA (ssDNA) was identified in 7 out of 30 cases.The positive rate was 23.3%,The point mutations were found in exons 7~8 of 2 cases.The results were influenced by many factors as well.It is indicated that strict experimental environment and condition is necessary to improvepositive rate and get right results of repetition.
